The Group A match also saw India pace spearhead Zaheer Khan playing in one of his scheduled domestic matches to assess his fitness to be able to join the Indian squad for the gruelling tour of Australia beginning next month.
He took one wicket, that of opener Bikas Pati, and conceded 20 runs from his six overs.
Mumbai resumed the day at 311 for three with Yadav on 69 and the batsman scored 200 from just 232 balls with the help of 28 fours and a six as his team declared their first innings at 529 for eight.
Yadav was the seventh Mumbai batsman out off Basant Mohanty but by that time his side had already reached a huge 515 runs.
Down the order, Dhawal Kulkarni contributed 61 runs and as soon as he was out in the post-lunch session, Mumbai declared their innings.
In reply, Orissa were staring at follow on as they were reduced to 57 for seven at stumps, a deficit of 472 runs with three first innings wicket in hand. The home side still need 321 more runs to avoid a follow-on, a tall task with the tailenders batting.
At stumps, Deepak Behera and Basant Mohanty were batting at 12 and seven respectively.
For Mumbai, Kulkarni took four wickets for 17 runs from his nine overs while Aavishkar Salvi bagged two wickets for five runs.
