"My desire is that the success in Yercaud constituency should be a trendsetter for AIADMK to emerge victorious in the Lok Sabha elections and to make revolutionary changes in India", she said in a statement.
The results of the by-poll should also signal an end to the Congress-DMK alliance, Jayalalithaa urged.
Meanwhile, District Collector and Returning officer for the election K Magarabhusnam said more than 2,500 police personnel would be deployed in 290 polling stations to ensure trouble free voting.
Magarabhusnam directed outsiders to leave the constituency ahead of voting.
Eleven candidates, including AIAMDK's P Saroja, wife of C Perumal, whose death necessitated the bye-election and V Maran of DMK will test their poll fortunes along with nine Independents.
